The real time polymerase chain reaction is a laboratory technique based on the polymerase chain reaction (PCR). While in Conventional PCR the DNA amplification is observed only as the end product of the reaction, in Real Time PCR the amplification of the DNA molecule can be monitored in real-time. Real time PCR can be used quantitatively (Quantitative real-time PCR), semi-quantitatively (Semi quantitative real-time PCR) or qualitatively (Qualitative real-time PCR). There are two most commonly spread methods for detecting the PCR products in real time PCR: A) Via non-specific fluorescent dyes that intercalate with any double-stranded DNA, B) Via DNA probes which are sequence-specific and are made of oligonucleotides labelled with a fluorescent reporter molecule(s). This method permits detection of PCR products only after the probe has hybridized with its complementary sequence. The Minimum Information for Publication of Quantitative Real-Time PCR Experiments (MIQE) guidelines suggest that the abbreviation qPCR should refer to quantitative real time PCR and that RT qPCR be used for reverse transcription–qPCR. The abbreviation “RT-PCR” commonly is used for reverse transcription polymerase chain reaction and not real-time PCR, however not all authors agree with this convention, therefore a careful examination of the terms should be done.

Storage Conditions: Store all reagents of the Acinetobacter Baumannii (Ab) Real Time PCR Kit at -20°C. Storage at +4°C is not recommended. All of the reagents in the kit can be used until the expiration date indicated on the box. Freeze and thaw cycles should be avoided as this will reduce the sensitivity of the kit. Up to 3 cycles of freezing and thawing will not influence significantly the performance of the kit. Cool all reagents during the working steps. Reaction mix should be stored in dark.
